(Color online) Schematic drawing of a gold coated AFM tip which is functionalized with oligo A. Upon approach of the tip toward the ds-DNA (oligo A∙oligo B) functionalized surface a triplex DNA can form. In the presence of target molecules, such as intercalating molecules, the stability of the triplex DNA is changed which can be detected by rupture of the third strand from the ds-DNA within a triplex DNA.

Force vs piezodisplacement curve recorded between an oligo A modified tip and a ds-DNA (oligo A∙oligo B) modified gold surface in buffer solution ( bis-tris buffer, 5.8, Spermine). Here at larger distances than no rupture event is observed and the signal represents the base line and the noise of the individual rupture force measurement. Only force peak values larger than were considered for further data analysis.
