An enlarged portion of the fiber, with six sequentially selected pixels, is used for polarization analysis, shown in (c) and (d). Based on the assumption of cylindrical symmetry, the long axes of the polarimetry plots (black arrows) reflect orientations of the underlying molecules. After decrimping, the arrows clearly become parallel, manifesting the reorientation of collagen molecules during the first stage of heating. Scale bar: .

Collagen denaturation quantified with SHG signal variation and shrinkage. Circularly polarized excitation was used. Temperatures of individual periods are marked at the top of figure. Three different stages (I, II, and III) are clearly distinguishable. The first stage, decrimping, occurs at , with slight shrinkage and unvaried SHG. In the second stage, remarkable shrinkage is observed with a concurrently decreased SHG. The rate of shrinkage and SHG decrease is similar during this stage. In the third stage with saturated shrinkage, SHG still decreases, reflecting an ongoing molecular uncoiling process.
