(Color online) (a) The presence of gradients in the scaffold libraries was determined by tracking the presence of Sudan IV dye in the scaffolds. Six gradients were cut into ten segments each, weighed, and dissolved in dioxane. The amount of Sudan IV in the dissolved segments was determined by measuring . Control clear and red scaffolds were also analyzed. for each scaffold segment was divided by mass to normalize for differences in sizes of segments. (b) “/mass” for six scaffold libraries was averaged and plotted against segment number. Clear (gray square) and red (red triangle) controls are given. Error bars are SD and the line is linear regression . (c) For cell culture, gradients were mounted in Teflon rigs in a Petri dish. The steel wire in scaffold enables mounting. (d) MC3T3-E1 cells were cultured for on libraries and fixed nuclei were stained to fluoresce green (SYTO-13 dye).
